The American biochemist Jennifer Doudna and French microbiologist Emmanuelle Charpentier (born 19 February 1964, Washington, D.C.), the French micronutrient Emmanuelle Charpentier, create a molecular tool known as the CRISPR-Cas9. The discovery of CRISPR-Cas9 in 2012 established the basis for gene editing, which let researchers to modify DNA sequences in a manner that was significantly more effective and technically simpler than previous methods. With the CRISPR-Cas9 technology, scientists were able to edit DNA to remedy genetic abnormalities in animals and to adjust the sequence of DNA in embryonic stem cells, which opened up the path to germline modification (sperm and egg) in humans. For their discovery and development of gene editing tools, Doudna and Charpentier have shared the 2020 Nobel Chemistry Prize.


In Hilo, Hawaii, Doudna spent a great deal of her youth. After graduating from Pomona College in California in chemistry in 1985, she moved to Harvard University. She was in the laboratory of the American biochemist and geneticist Jack W. Szostak (who was awarded the 2009 Nobel Prize for Physiology or Medicine) and graduated in biochemistry in 1989. She joined the Yale University faculty in 1994 after doing post-graduate studies at the University of Colorado under the guidance of American biochemist and molecular biochemist Thomas R. Cech (who earned a share of 1989 Nobel Chemistry Prize). In 2002 she relocated to Berkeley University, where she taught biochemistry and molecular biology.

She later studied genetic control by several short RNAs and became interested in CRISPR. CRISPR is part of the immunological system of the bacteria. It comes from RNA sequences from invading viruses integrated into bacterial genomes. The viral sequences exist in the spaces between short blocks of bacterial DNA sequences as DNA. The virus will convert the spacer DNA to RNA the next time it invades a bacterial cell. The enzyme Cas9 and another RNA molecule join the newly encoded RNA which then looks to match viral DNA strands. When found, Cas9 breaks viral DNA to limit replication of the virus. Doudna and Charpentier found that Cas9 can be altered to an exact DNA sequence in the RNA guidelines. Their discovery quickly altered the genome engineering environment, giving new options for human illness treatment.

Human genome engineering was an unavoidable byproduct of rapid developments in technology of genetic engineering. However, there has been insufficient knowledge concerning the safety of genetic engineering and its application to edit human DNA, specifically as regards whether genetic engineering technology should be utilised to enhance non-disease features, like intelligence. Doudna launched a campaign in early 2015 for a moratorium on human genome editing and set up a framework of immediate action in April of that year to save human embryonic genomes from change. However, in spite of cautious efforts, Chinese scientists reported altering human embryo genomes with CRISPR-Cas9 in April 2015.

In addition to the Nobel Prize, Doudna received other honours for her research, including the Gruber Prize for Genetics (2015) and the International Canada Gairdner Award (2016), which were shared with Charpentier. Doudna was an elected member of several academies and investigator of the Howard Hughes Medical Institute (from 1997).
