David Sinclair Biography and Career:

David Andrew Sinclair AO, a genetics professor and co-director of the Paul F. Glenn Center for Biology of Aging Research at Harvard Medical School, was born in Australia on June 26, 1969. He is renowned for his studies on epigenetics and ageing. He holds the Australian Order of Merit (AO).Sinclair has made appearances in publications such as The New York Times, Boston Magazine, The Washington Post, The Economist, TED, The Joe Rogan Experience, The Charlie Rose Show, and 60 Minutes.

David Andrew Sinclair was raised in the New South Wales town of St Ives after being born in Australia in 1969. After the 1956 Hungarian Uprising was put down, his paternal grandmother immigrated to Australia, and his father changed the family name from Szigeti to Sinclair. In addition to winning the Australian Commonwealth Prize, Sinclair graduated with a bachelor’s degree in science from the University of New South Wales in Sydney.

The same university awarded him a Ph.D. in molecular genetics in 1995with a focus on yeast gene regulation. When Massachusetts Institute of Technology professor Leonard P. Guarente visited Australia in 1993 for a lecture tour, Sinclair had the opportunity to meet him. This encounter inspired Sinclair to apply for a post-doctoral position in Guarente’s lab. A single gene mutation in (Daf-2) had been found to be capable of doubling the lifespan of C. elegans earlier that year by Cynthia Kenyon’s team at UCSF.

At Harvard Medical School, Sinclair was hired in 1999.In 2003, his lab was insufficiently funded and small. In 2004, Sinclair met with philanthropist Paul F. Glenn, who gave Harvard $5 million to start the Paul F. Glenn Laboratories for the Biological Mechanisms of Aging, of which Sinclair was appointed the founding director. He and Bruce Yankner are the co-directors at the moment.

Sirtris Pharmaceuticals was founded in 2004 by Sinclair, serial entrepreneur Andrew Perlman, Christoph Westphal, Richard Aldrich, Richard Pops, and Paul Schimmel. Sirtris concentrated on advancing Sinclair’s investigation into sirtuin activators, which had its origins in the Guarente lab.The company was focused specifically on resveratrol formulations and derivatives as activators of the SIRT1 enzyme; Sinclair gained notoriety for describing resveratrol as “as close to a miraculous molecule as you can find.”

Higgins on the use of T cell-stimulating antigens to develop vaccines, Genocea Biosciences was founded in 2006; Sinclair was a co-founder.Sinclair received a promotion to tenured professor at Harvard Medical School in 2008. He also accepted a joint professorship at the University of New South Wales’ School of Medical Sciences a few years later.

When the Wall Street Journal asked for an interview with Sinclair about his work with the company and its marketing, he disputed the use of his name and words to promote the supplement and resigned. Sinclair joined the scientific advisory board of Shaklee in 2008 and assisted them in developing and introducing a resveratrol-containing product called “Vivix.”Based on research conducted by Tilly on mammalian oogonial stem cells and Sinclair’s work on mitochondria, OvaScience was founded in 2011 by Sinclair, Michelle Dipp (who had previously worked with Sirtris), Aldrich, Westphal, and Tilly.

Some groups were unable to duplicate Tilly’s work, which made it controversial. Along with Nir Barzilai and other colleagues, Sinclair also co-founded CohBar in 2011. New peptides derived from mitochondria were to be found and developed as part of CohBar. In 2015, Sinclair talked about his efforts to raise money for his lab in an interview with The Scientist. He explained how his lab expanded to about 20 people, then shrank back down to about 5, before growing once more as he received funding from philanthropic organisations and businesses, including ones that he helped start.

His lab employed 22 people in 2015 and received one R01 grant, with non-federal funding accounting for 75% of its funding.  By 2016, however, his federal funding had started to rise, so this was no longer the case. Co-written with journalist Matthew LaPlante and released by Sinclair in September 2019 as a New York Times bestseller, Lifespan: Why We Age – and Why We Don’t Have To.

Sinclair narrated the audiobook version of this, which was also published on Audible. Sirtuin 1 (also known as sir2 in yeast) slows the ageing process in yeast by lowering the buildup of extrachromosomal rDNA circles, Sinclair discovered while working in Guarente’s lab. NAD was recognised as a crucial cofactor for sirtuin function at the time by other lab workers.He and Guarente had a disagreement in 2002 at a scientific gathering at Cold Spring Harbor Laboratory after Guarente had left for Harvard.

He disputed Guarante’s explanation of how sir2 might be involved in ageing, which sparked a scientific rivalry. When Sinclair’s lab was still small, in 2003, he learned that researchers at the Pennsylvania biotech company Biomol Research Laboratories had discovered that polyphenols like resveratrol could activate sir2. He worked with them to confirm this.As a result, Sinclair contributed to publications in both Nature and Science in 2003.

Sinclair’s vociferous promotion of resveratrol as an anti-aging substance sparked a scientific debate over whether this was accurate and whether resveratrol even activated sirtuins.Also under close examination are prominent papers that assert age reversal in mice. Mice given high doses of resveratrol had more mitochondria in their cells, according to research from another lab that was partially funded by Sirtris.

In order to better understand ageing and develop strategies to delay it, Sinclair’s lab continued to investigate resveratrol and its analogues, as well as mitochondria and NAD. Sinclair has won numerous honours for his work, including the Australian Society for Medical Research Medal in 2014, the American Federation for Aging Research Irving S. Wright Award of Distinction in 2018, and the Australian Government Advance Award in Life Sciences in 2017.

Sinclair was listed as one of the 100 most influential people in the world in Time 100 in 2014, and he was listed among Time magazine’s 50 Most Influential People in Healthcare in 2018. Sinclair received the Order of Australia (AO) in 2018 in recognition of his “distinguished service to medical research into the biology of ageing and lifespan extension, as a geneticist and academic, to biosecurity initiatives, and as an advocate for the study of science” (2018 Australia Day Honours)
