Feng Zhang Biography and Career:

Feng Zhang is a Professor of Brain and Cognitive Sciences and Biological Engineering. Zhang is not just a researcher at Harvard but also at the Howard Hughes Medical Institute. Zhang is a molecular biologist who specializes in the development and use of innovative molecular technologies for the study of the brain.

Zhang pioneered the invention of tools for editing the genome that can be used in eukaryotic cells, such as human cells, using CRISPR-Cas9 systems naturally found in microbes. He and his colleagues have modified several different CRISPR systems to be utilized as tools for genome engineering. These systems include RNA-targeting CRISPR-Cas13 systems and CRISPR-associated transposon systems, which can be used to insert genes.

Zhang uses CRISPR and other approaches to investigate the genetics and epigenetics of human illnesses, particularly complex disorders like psychiatric and neurological diseases. These diseases are brought on by a combination of genetic and environmental risk factors, and they are relatively easy to model using methods that are more traditionally used.

The tools that he developed in his lab and made publicly accessible are being used in various research areas, including immunology, clinical medicine, and cancer biology, to name a few. His long-term objective is to create innovative therapeutic approaches for treating various diseases.

The Zhang lab is dedicated to the study of neurobiology via the application of synthetic biology to develop technologies for genome and epigenome editing. He is considered a pioneer in optogenetics, which was recognized as the “Method of the Year” in 2010. He started working on employing TAL effectors to regulate gene transcription as a postdoc.

Early in 2011, Zhang started efforts to harness and optimize the CRISPR system so that it might function in human cells, based on work on prior research conducted at the Sylvain Moineau Lab. Emmanuelle Charpentier and Jennifer Doudna’s groups worked together to describe a chimeric RNA design capable of aiding DNA cleavage using purified Cas9 protein and a synthetic guide.

When Zhang’s group was working on optimizing the Cas9 system in human cells, the collaborating groups of Jennifer Doudna and Emmanuelle Charpentier reported the design. Comparing their method of RNA production with a plan based on the Doudna / Charpentier chimeric RNA for usage in human cells, the Zhang group identified aspects of the guide essential for Cas9 to work successfully in mammalian cells.

These qualities are not required for Cas9 to function effectively in biochemical experiments. In September 2013, Zhang, Jennifer Doudna, and a few other Harvard colleagues established Editas Medicine to develop and commercialize CRISPR-based medicines.

Zhang and his Harvard colleague Eugene Koonin used computational biology approaches to find Cas13. In 2016, Zhang helped to establish Arbour Biotechnologies to develop Cas13 for medical applications. His group has also created a sensitive diagnostic nucleic acid detection process called SHERLOCK based on CRISPR. This protocol can identify and differentiate strains of viruses and bacteria present in concentrations as low as attomolar (1018 M).

In 2018, Zhang co-founded Sherlock Biosciences, which focuses on the continued development of diagnostic technologies. Additionally, in 2018, Zhang collaborated with David R. Liu, the creator of Editas and a Harvard colleague, to establish Beam Therapeutics to advance Liu’s work on base editing and prime editing. According to Google Scholar, his h-index is 109. He has published 109 articles.

Zhang was selected as a Searle Scholar in 2012 and received the NIH Director’s Pioneer Award. In 2013, MIT Technology Reviews recognized him as one of their TR35 innovators. In addition, he was awarded the 2014 Young Investigator Award by the Society for Neuroscience. Zhang was also awarded the Robertson Stem Cell Investigator Award by the New York Stem Cell Foundation (NYSCF) in 2014.

The same organization recognized him as the 2016 Robertson Stem Cell Prize recipient. In 2015, Zhang was presented with the Tsuneko & Reiji Okazaki Award for the first time. The following year, in 2016, he received the Gairdner Foundation International Award and the Tang Prize. He had the honor of winning with Doudna and Charpentier for the second and third times.

The Albany Medical Centre Prize and the Lemelson-MIT Prize were bestowed to him in the same year (2017). The Technion awarded him the Harvey Prize in Israel for 2018, which he earned in 2019. 2019 was the year that Zhang was presented with the Golden Plate Award from the American Academy of Achievement. The Richard Lounsbery Award was bestowed to him the following year, in 2021.

Zhang was given membership into the National Academy of Medicine, the National Academy of Science, and the American Academy of Arts and Sciences in 2018. Zhang is also a Fellow of the American Academy of Arts and Sciences.
