John O’Keefe Biography and Career:

John O’Keefe, FRS FMedSci, is a neuroscientist, psychologist, and Professor at the Sainsbury Wellcome Centre for Neural Circuits and Behaviour and the Research Department of Cell and Developmental Biology at the University College London. He was born in the United States on November 18, 1939, and has dual citizenship in both countries.

He found that the hippocampus comprises “place cells” and exhibits theta phase precession, a sort of temporal coding. He has won several accolades, including the Nobel Prize in Physiology or Medicine, which he shared with May-Britt and Edvard Moser in 2014. His whole career has been spent at University College London, albeit at the demand of his Norwegian coworkers, the Mosers; he also held a part-time position at the Norwegian University of Science and Technology.

O’Keefe, born in New York City to Irish immigrants, graduated from Regis High School (Manhattan) and the City University of New York with a BA. Later, he attended McGill University in Montreal, Quebec, Canada, where he studied under Ronald Melzack and earned an MA in 1964 and a Ph.D. in Psychology in 1967.

In 1967, O’Keefe traveled to UCL as a postdoctoral research fellow from the US National Institute of Mental Health to study under the late Patrick Wall. He was appointed as Professor in 1987 and has remained there ever since. In 2014, he was given a part-time professorship at the Norwegian University of Science and Technology at the urging of his coworkers Edvard Moser and May-Britt Moser.

Through a methodical examination of the effects of context on the activity of single hippocampus neurons, O’Keefe and his student Jonathan Dostrovsky identified place cells. The numerous articles he’s written on place cells are often referenced. He produced a seminal book with Lynn Nadel outlining the hippocampus’s role as a cognitive map for spatial memory. Hundreds of studies have extended his work by doing experimental analysis of place cells or modeling their behavior.

O’Keefe discovered in his follow-up study of place cells that the timing of action potentials in place cells differs from the timing of spikes inside a single cell, suggesting that this information is coded temporally in a manner separate from that used by other types of neurons. Together with Michael Recce, he published research in 1993 showing that place cells spike at different times about the theta rhythm oscillations in the hippocampus’ local field potential.

When a rat first enters a place cell’s firing field, the spikes occur at the end of the theta rhythm, but as the rat continues to walk through the area, the points move to the beginning of the cycle. Multiple following publications have shown similar results, supporting the idea that spike timing is used to encode sensory data. The probable physiological processes of theta phase precession have been the subject of several models.

In a 1996 publication, O’Keefe and Neil Burgess showed that when the boundaries defining the environment were changed, the location and size of the firing fields of place cells also changed. They proposed a model for this phenomenon in this and later articles, which predicted the presence of boundary vector cells that would react at a certain distance from environmental boundaries.

Extensive experimental evidence indicating boundary cells with the anticipated features in the subiculum and the medial entorhinal cortex (where they are often referred to as border cells) emerged some years later to provide weight to this explicit theoretical prediction.

O’Keefe was made a member of the Academy of Medical Sciences in 1998 and the Royal Society in 1992. He also won the Grawemeyer Award in psychology in 2006 (jointly with Lynn Nadel) and the Feldberg Foundation Prize in 2001. Later the same year (2008), O’Keefe received the Gruber Prize in Neuroscience. He is the center’s first-ever director, thanks to his appointment as such by the Sainsbury Wellcome Trust.

The Louisa Gross Horwitz Prize was awarded to him in 2013 (together with Edvard Moser and May-Britt Moser). He shared the 2014 Kavli Prize with Brenda Milner and Marcus Raichle from the Norwegian Academy of Science and Letters. The National Academy of Sciences accepted him as a member in 2016. He became an honorary member of the Royal Irish Academy in 2019.

On December 15, 2014, University College Cork awarded O’Keefe an honorary Doctor of Science degree. His alma institution, The City College of New York, presented him with one in May 2015, while his alma mater, McGill University, presented him with one in June of the same year. O’Keefe appeared on the March 10, 2015, episode of The Life Scientific on BBC Radio 4.
