James P. Allison Biography and Career:

James P. Allison, Ph.D., holds the Vivian Smith Distinguished Head in Immunology, is a professor and chair of the Department of Immunology, and is the Executive Director of the immunotherapy platform.

Allison’s research is focused on understanding the processes that regulate T-cell responses and putting that fundamental knowledge to use in the fight against cancer by overcoming its ability to evade the immune system’s assault. His foundational findings led him to pioneer the use of immune checkpoint inhibition as a therapy for cancer.

He collaborated with pharmaceutical firms to create Yervoy, which was authorized by the Food and Drug Administration of the United States in 2011 for treating metastatic melanoma. He continues his work in fundamental research and translational trials of immunotherapies and combination treatments to discover the parameters determining patient response and resistance to checkpoint blockage.

Both the National Academy of Medicine and the National Academy of Sciences count Allison as a member in good standing. Several prestigious prizes and accolades have been bestowed upon him, including the Lasker-DeBakey Clinical Medical Research Award in 2015 and the Breakthrough Prize in Life Sciences in 2014.

Allison’s participation in a scientific-training program at the University of Texas at Austin (UT Austin), when he was 15 years old contributed to his interest in science, which had been building for some time. Subsequently, he attended the University of Texas at Austin, receiving a Bachelor of Science degree in microbiology in 1969 and a Doctor of Philosophy degree in biological sciences in 1973.

After that, he did a postdoctoral fellowship at the Scripps Clinic and Research Foundation in La Jolla, California, which was renamed Scripps Research Institute. Allison spent most of her time at Scripps working on amino acid sequencing, but she also managed to squeeze in time to research immune cells’ functions. The results of these trials led to his first significant discovery, which concerned how the immune system recognizes tumors.

Allison relocated to the University of Texas MD Anderson Cancer Centre in the middle of the 1970s and began working as an assistant scientist there shortly after her arrival. His study was based on discovering how T cells may recognize foreign particles, sometimes known as antigens.

He also accomplished a successful mapping of the structure of the T-cell antigen receptor during this period. Allison decided to take a full professorship in the field of immunology at the University of California, Berkeley, in 1985. Before that, she had worked in various capacities at the University of Texas at Austin, Stanford University, and MD Anderson.

In that institution’s laboratory, he discovered CD28, which turned out to be an essential costimulatory signaling molecule critical for the activation of T cells. Additionally, he and his coworkers found that a molecule known as CTLA-4 acted as an antagonist to CD28 and played an essential part in downregulating immune responses. CTLA-4 antibody, intended to prevent the function of CTLA-4, improved T-cell responses and reduced the size of tumors in mice treated with the antibody.

Immune checkpoint blockade is the name given to this strategy for enhancing immune responses over time. In his studies, Allison progressively concentrated on developing a deeper understanding of the impacts of CTLA-4 inhibition.

Amid Allison’s research on CTLA-4 inhibition, he relocated from Berkeley to Weill Cornell Medical College in New York City. There, he worked concurrently as the head of the Ludwig Centre for Cancer Immunotherapy at the Memorial Sloan Kettering Cancer Centre from 2006 until 2012. While there, he collaborated with a pharmaceutical business to create a human monoclonal antibody against CTLA-4 called ipilimumab that could be administered to cancer patients.

In 2011, the antibody became the first immune checkpoint medication authorized by the Food and Drug Administration of the United States; it was approved for treating advanced stages of melanoma and was marketed under the brand name Yervoy.

In 2012, Allison returned to work at the MD Anderson Cancer Centre, where he previously held positions as the Director of the Parker Institute for Cancer Immunotherapy and the Vivian L. Smith Distinguished Chair in Immunology. Allison was the winner of many accolades, including the Nobel Prize, the Canada Gairdner International Award (2014), and the Louisa Gross Horwitz Prize (2014), all of which she received in 2014.

He was elected to many organizations, some of which include the National Academy of Medicine (2007), the National Academy of Sciences (1997), and the American Association for the Advancement of Science (2006).
